Title: “Quantitative analyses of growth arrested bacteria the evolution of antibiotic tolerance”

Abstract: Since the pioneering work of Jacques Monod who laid the foundation for quantitative studies of growing bacteria, most microbiological studies have focused on exponentially growing bacteria. However, in the wild, microorganisms tend to spend most of their lifetime at stationary phase in a growth-arrested state. Despite this general prevalence growth-arrested bacteria and their role in bacterial persistence, they are as yet poorly characterized. We develop new tools for the analysis of single growth arrested bacteria. Surprisingly, we identify conditions in which single bacteria produce proteins over many days at a constant rate, despite their growth arrest, which we term CASP (Constant Activity at Stationary Phase). Finally, we dissect a pathway for controlling growth arrest and show its involvement in the evolution of antibiotic tolerance.
